Abstract: Σκοπός της παρούσας εργασίας είναι η διερεύνηση και βέλτιστη παραμετροποίηση ενός μοντέλου πρόβλεψης της εμφάνισης δυναμικών δράσης βάσει των δυναμικών τοπικού πεδίου της περιοχής του υποθαλαμικού πυρήνα ασθενών που πάσχουν από την νόσο του Πάρκινσον. Τα δεδομένα που χρησιμοποιούνται και αναλύονται παραχωρήθηκαν από την νευροχειρουργική ομάδα του κ.Δαμιανού Σακκά και αποτελούν μικροηλεκτροδιακές καταγραφές της περιοχής του υποθαλαμικού πυρήνα τριών ασθενών κατά τη διάρκεια της νευροχειρουργικής επέμβασης για την εν τω βάθει διέγερση του συγκεκριμένου πυρήνα (Deep Brain Stimulation-DBS).Συγκεκριμένα, γίνεται μελέτη του σύνθετου σήματος της εξωκυττάριας καταγραφής, παρουσιάζονται οι συνιστώσες του και εξάγονται αυτές που θεωρείται ότι συνδέονται με σχέση εισόδου-εξόδου στην περιοχή του πυρήνα. Στη συνέχεια επιχειρείται η μοντελοποίηση της σχέσης χρησιμοποιώντας τμήματα των ληφθέντων σημάτων για την εκπαίδευση ενός μη γραμμικού μη παραμετρικού μοντέλου μαύρου κουτιού Hammerstein-Wiener (H-W). Συγκρίνοντας τον βαθμό ομοιότητας της εξόδου των μοντέλων και των πραγματικών καταγραφών αξιολογούμε και επιλέγουμε τα καλύτερα. Η μεθοδολογία αυτή μπορεί να γίνει οδηγός για την ψηφιοποίηση και μεταφορά σε υπολογιστή των δεδομένων που προκύπτουν από τον υποθαλαμικό πυρήνα και μοντελοποίησή του. Μοντέλα αυτού του προσανατολισμού μπορούν να χρησιμοποιηθούν σαν πλατφόρμες δοκιμών για τη βελτιστοποίηση των παραμέτρων της νευροδιέγερσης. The scope of this thesis is the creation of a model of the spiking activity taking place inside the subthalamic nucleus (STN) based on the local field potentials. The spiking activity recorded from the STN of patients suffering from Parkinson’s disease is simulated using a Hammerstein-Wiener model. The data that were used are recordings from the area of the STN of the patients at the time of implantation of the electrodes for deep brain simulation (DBS) provided by Professor Damianos Sakas neurosurgeons’ group. Specifically, the extracellular recording and its components are brought into focus and analysis and we assume an input-output relationship between its filtered components. Furthermore, using segments of the recorded data this thesis tries to model this relationship using black-box non parametric non linear modeling. Moreover, it assesses how well the proposed cascade model fits to the measured data and proposes the best one for each recording.In conclusion, the thesis contends an excellent agreement between the predicted data according to the H-W model and the measured one. Further research could prove its predictability to be accurate for other recordings too, thus establishing its validity as a simple yet biologically plausible model of the spiking activity recorded from the motor area of the subthalamic nucleus of Parkinson’s disease patients.
